[Much binned] You say "mixer" -- do you mean alsamixer? If not then run alsamixer as user on a command line and see (using F6) that there is a card available. (You haven't selected the card on the mobo. which you have deactivated in the Bios? Just wondering...)

Aaah, that pulseaudio ... love it or hate it, it will be there... Don't forget pulseaudio sits ontop of alsa so do the alsamixer thing. Oh, when you click on the Speaker (bottom right on taskbar --ie, if you do have this speaker) is the volume set to more than 0? BC -- God created war so that Americans can learn geography.
