Sometimes, after a new installation or an update or something that changes my mixer settings, my mic refuses to work. You would think that the standard response would be to make sure the mic is turned up and not muted. However, if there is more than one mic input, alsa doesn't always make a logical assumption of which one should be default. Even after changing that, I have to make sure capture is not muted. Unmuting capture on my main machine gives me a terrible squeal, so I futz around until I get rid of that (never having really put any mental effort into it, I couldn't tell you how I did that).
In addition, sometimes software has its own rules about what it takes sound input from. For instance, if you use KDE4 and want to make sure that's working correctly, you need to look at the sound devices it uses.
