I've use a GNOME desktop since the Ximian days and never seen this issue. If I run "update-mime-database /usr/share/mime" it completes without error. Did you install any packages, etc... before this broke? [Maybe KDE4 or vlc?] If you update with the "-v" option does it say anything interesting? A quick Bing search finds a couple posts relating this to KDE4 packages. Do you have KDE4 installed?
