So you just mean that the clock should show seconds? Well, Plasma's clock has an explicit setting for this, no need to choose a locale that includes the seconds in the "short format" (though you might prefer it for other applications of course). Actually, whether the clock displays seconds or not is independent of the used locale, except for this bug of course (that got introduced when adding the 24h option in 5.4 btw, it worked fine before up to 5.3.3).
I can't see how in whatever version of whatever the clock is part of in KF 5.20.0 and QT 5.5.1 in TW20160408. Date format is not a selectable field when en_US is selected region.
There is a setting for the displayed "Date format" in the "Digital Clock Settings" (the last one at the bottom). You can choose between "Long Date", "Short Date", and, ... "ISO Date". That option was introduced in Plasma 5.4 (like the explicit 24h option) and it's independent of the locale in use (actually it was introduced as a workaround for the complaints of not being able to easily configure/use the ISO date format in KDE any more). This option is only selectable if "Show date" is enabled though, it doesn't make sense otherwise as it only affects the digital clock.
