I don't have all the answers for you, but grub, at least, has to be on the master drive, a bootable floppy, or a bootable cd/dvd. You have more flexibility about where the second stage image is, and where the root filesystem is located.
As for having the root filesystem on a USB drive, that might be possible, but I would think not until after the kernel is loaded so the system has access to USB devices.

Hello, If your computer is able as mine, to boot from a USB drive, you can install GRUB on the MBR of this drive. It works for me. Michel.
