Short answer "Yes". Do it in run level 1 or 3 but avoid doing this in run level 5 or GUI mode. Too many GUI programs depend on whats in /tmp or /var/tmp and won't operate correctly until you logout/login again and those files get rebuilt. I always prefer to do this in single-user mode (run level 1). There are daemons that may use /tmp.
But, in Linux, when you "delete" a file, that file does not really go away.
The rm(1) command causes the use count to decrement. If that file is
currently being used, it will not be physically deleted.
