Edit /etc/X11/XF86Config (yes, I'm sure it can be done in a GUI too) and find the section "InputDevice" that deals with the mouse. Make sure it has a line that reads Option "ButtonNumber" "5" and one that reads Option "ZAxisMapping" "4 5" then restart X and see if it helps
