=> not a CUPS issue but a general issue regarding the serial port.

CUPS autodetects, what can be autodetected
in the current system.
CUPS does not change the underlying system.

I.e. first it must work to send data via the serial port
to the printer similar as described in
http://en.opensuse.org/SDB:Installing_a_Printer
e.g. by using as root command like
# echo -en "\rHello\r\f" >/dev/ttyS0
(replace '/dev/ttyS0' with the device node for your serial port).
