If you are not using GNOME, try to remove /etc/asound-pulse.conf if exists.
If you still have the problem, please elaborate what you tested: i.e. which
application doesn't work in which situation. As a simpler testing, run a
standard program like aplay on a terminal:
% aplay -vv somefile.wav
